Isolation of Lower Molecular Protein Doublet from Human Stratum Corneum


Abstract
The biochemical properties and immunohistochemical locailzation of 14.4Kd and 17Kd protein of normal human stratum corneum were investigated. This protein doublet was isolated from human stratum corneum by Sephadex G 50 column and reversed phase HPLC. Each amino acid composition of 14.4Kd and 17Kd protein is quite similar. Serine and glycine are rich, but proline and half cystine are poor.
Rabbit polyclonal antibody to 17Kd protein extracted in 8M urea reacts with 17Kd as well as 14.4Kd extracted in urea and PBS both. Immunohistrochemical studies show that localization of this protein doublet changes from suprabasal cytoplasm of living epidermal cell layer to the cell membrane of corneocyte. These results suggest that this protein doublet is a quite different from well known matrix proteins of epidermis. Therefore, it seems to be a unique soluble matrix protein of epidermis and a new marker protein of epidermal differentiation.
